内容記述 Histamine causes allergic symptoms. It is synthesized in mast cells by decarboxylation of histidine by histidine decarboxylase (HDC). As histidine is an essential amino acid, the human body absorbs it through dietary sources, and a diet containing excess histidine may affect histamine synthesis. The researchers have previously shown that a high concentration of histidine promotes degranulation in RBL-2H3 cells. However, the transport pathway of histidine to mast cells is currently unclear. Therefore, the expression of genes encoding histidine transporters in RBL2H3 cells was assessed, in order to clarify the histidine transport pathway in mast cells and the effect of histidine on degranulation. After transporting histidine into the mast cells, L-alanyl-L-alanine which is a competitive inhibitor of histidine was utilized to induce degranulation. The induction of degranulation was examined by measuring β-hexosaminidase release rate.
The expression of histidine transporters' genes, PHT1, PHT2, and PEPT1 in RBL-2H3 cells was analyzed by RT-PCR. The release rate of β-hexosaminidase was measured by adding histidine at final concentrations of 0.5 mM to 1.0 mM and 50 mM L-alanyl-L-alanine to RBL-2H3 cells. After incubating the cells for 24 hours, they were stimulated with the calcium ionophore A23187 to induce degranulation.
PHT1 gene expression, which transports histidine, dipeptide, and tripeptide, was observed, but neither PHT2 or PEPT1 expressions were observed. The rate of β-hexosaminidase release was increased by the addition of 0.5 mM and 1.0 mM histidine, which enhanced degranulation induction. In contrast, simultaneous addition of both histidine and 50 mM L-alanyl-L-alanine showed a significant decrease in β-hexosaminidase release rate compared to addition of histidine alone. This suggests that L-alanyl-L-alanine might have inhibited the histidine uptake pathway, thereby suppressing the induction of degranulation.
Therefore, this study shows that histidine is transported into the mast cells via PHT1 and L-alanyl-L-alanine inhibits histidine transport into the mast cells. Future study is needed to further clarify the histidine transport pathway into the cell and its effect on histamine synthesis.
